Mechanism of the Oxidation of Tyrosine by Amino Acid Oxidase of Snake Venoms †

چکیده
It has been known since the studies of Neubauer in 1909 and of Knoop in 19107"' that the mammalian organism is able to degrade a-amino acids to a-keto acids. Knoop advanced the hypothesis that this degradation involved an oxidative deamination.' According to this hypothesis, the imino derivative of the amino acid is first formed in an oxidative step and, in turn, the imino compound yields the corresponding keto acid in an hydrolytic step. The imino acid intermediate, according to Knoop, may be in equilibrium with its hydrated form:

l-Amino acid oxidase isolated from Calloselasma rhodostoma snake venom induces cytotoxicity and apoptosis in JAK2V617F-positive cell lines
BACKGROUND Myeloproliferative neoplasms are Philadelphia chromosome-negative diseases characterized by hyperproliferation of mature myeloid cells, associated or not with the Janus kinase 2 tyrosine kinase mutation, JAK2V617F. As there is no curative therapy, researchers have been investigating new drugs to treat myeloproliferative neoplasms, including l-amino acid oxidase from Calloselasma rhod...
متن کاملIsolation and Purification of L-amino Acid Oxidase from Indian Cobra Naja naja
The enzyme L-Amino Acid Oxidase (LAAO) is widely distributed in snake venoms, contributes a reasonable toxicity to the venoms. However LAAO from Indian cobra (Naja naja ) snake venom has not yet isolated previously. In the present study LAAO from Indian cobra (Naja naja ) snake venom was purified by sequential steps of cation exchange chromatography (CM-Sephadex C-25; CM-52 cellulose), followed...
متن کاملSnake venom proteins and coagulopathy caused by snakebite
Snakebite affects around 3 or 4 million humans annually leading to more than 100,000 deaths. Coagulopathy is one of the significant causes of both morbidity and mortality in these patients. Accordingly, it is of utmost importance to diagnose and treat coagulation disorder due to bites; in addition, it is accompanied by various clinical aspects, such as pre-coagulation, fibrinogen coagulation ti...
متن کاملMathematical modelling of amino acid resolution catalyzed by L-amino acid oxidases from Crotalus adamanteus and Crotalus atrox
Amino acid resolutionwas studied in this paper. L-Methionine and DL-methioninewere used as substr for the two snake venom L-amino acid oxidases (from Crotalus adamanteus and Crotalus atrox). E though these enzymes have similar background, they exhibit differences as well as similarities. It found that both enzymes are active towards L-methionine as substrate (Vm = 0.78 mmol min 1 g ) have simil...
متن کاملQM/MM Study on the Mechanism of Aminophenol Oxidation by Functionalized β-Cyclodextrin as Oxidase Nanomimic
In this study, functionalized β-cyclodextrin (β-CD) by aldehyde group was investigated as an oxidase enzyme mimic for the amino phenol oxidation. All calculations were performed by GAUSSIAN 09 package using two layers ONIOM method at the ONIOM (MPW1PW91/6-311++G(d,p)/UFF) level. In the first step, H2O2 is encapsulated in the hydrophobic cavity. In the second step, H2<...
